NKX3.1 has been identified as a marker for metastatic tumors. NKX3.1 is positive for prostate staining cells with nuclear staining. Most cases of urothelial carcinoma were found negative for NKX3.1, allowing to distinguish high-grade prostate adenocarcinomas and high-grade infiltrated urothelial carcinomas. NKX3.1 is also expressed in invasive ductal carcinomas (IDC) and breast invasive lobular carcinomas (ILC).
NKX3.1 expression is limited to ER, PR and AR positive carcinomas, and is more encountered in ILCs than in IDCs.
